An immune repertoire is the sum of functionally diverse T and B cells. It provides both a snapshot and a historical record of immune function — a powerful tool for characterizing the adaptive immune system’s responses to cancer, autoimmune and infectious diseases, allergies, vaccinations, and therapeutic treatments.

Profiling the TCR and BCR variable regions using RT-PCR and NGS provides critical data for the discovery of novel, disease-associated immunity biomarkers. Both qualitative changes (disease-specific CDR3s) and quantitative changes (repertoire diversity) can be captured.

The CDR3 region, located at the junction of V, D, and J gene segments, shows the greatest variability. It can further increase diversity through V(D)J rearrangements, nucleotide insertions/deletions, and somatic hypermutation after antigen stimulation.
